Hello,

On a 12 marker test from FTDNA two of our Johnston subjects had 12 at DYS 388
and one had 10 at 388. According to Bruce Walsh's table using the high
mutation rate this could occure somewhere between 3 - 61 generations. We are
about 4 transmission events from our common ancestor. Can anyone tell me
where to look for mutation frequency for DYS 388?

Because of a known genealogy back to the g-g-grandmother we are certain of
our relationship. Unfortunately, the 11/12 results on the Y test fails to
confirm that both her sons had the same father. I'm inclined to believe we
have a two step mutation at 388 -- and a common ancestor -- but I really
would like to know what others have found at that locus.

If two step mutations at 388 are common I think we'll just assume one
happened in our family. However, if mutations at DYS 388 are rare or if no
two step mutations have been reported there, upgrading to a 25 marker test
seems to be our next step.
