Yes, you can do that.

By "before" you mean their ancestors, or ??? Are they to be included

You could use the Descendant Interest flag, but I'd create a new flag
for the purpose, with values of N,Y. The main issue is exactly who do
you want to include in the "descendants?" Only the actual descendants,
or also their spouses? How about parents of the spouses? Other spouses
of those spouses? Children by their "other spouses?" How about
unrelated witnesses to events any of the above are linked to?
Any of these are possible, but you have to be clear exactly who you
want to include to get the results you expect. Once you decide that,
and choose either the existing Descendant Interest flag or create a
new one, you set the flag to the assigned value for those you want to
include, then use the List of People report with a filter for that
flag and value, and a Secondary output of New Project to create the
new project.
If the number of descendants involved is quite small you could set the
flag manually. But if there are very many you will want to use the
List of People report and it's Secondary output to change the flags.

provide more specific details.
One thought -- there's a bug in the current release with using custom
flags with filters, so perhaps using the Descendant Interest flag
would be a better choice until that's fixed.
